The CRIB standard protocol for lipidomics follows a novel chromatography strategy involving methanol and isopropanol as the mobile phase using an Agilent Poroshell-120 EC-C18 column. This ESI-LC-MS protocol was developed in the Agilent Center of Excellence in Delaware by Sumit Shaw. The protocol leverages the excellent separation of lipid classes along with accurate mass detection and high fidelity transmission of isotopic envelopes. Two injections are performed of the sample, one in positive ion mode and one in negative ion mode.

A targeted lipid assay for signaling lipids is available (see targeted metabolomics), and this chromatography method is also available depending on the classes of lipids being measured.
